Layer by Layer coated CaCO3 microparticles for oral immunization of Hepatitis B antigen

Abstract

Oral delivery of peptide is most affordable delivery route for delivery of bioactive agents because it provides freedom from the side effects caused by needle in (intramuscular) IM or (intra-venous) IV administration. Oral delivery of peptides is quite difficult because of the presence of proteolytic enzymes in gastrointestinal tract (GIT) that breaks the peptide in small segments and resulted in the loss of activity. In the present study we have developed Layer by Layer (LBL) coated CaCO3 microparticles for the delivery of HBsAg via oral administration. Formulated microparticles were of 4.15±0.35µm in size and zeta potential of -32.4±3.69mV. LBL coated microparticles shows sustained release of HBsAg than non coated MPs. In-vitro cell uptake and in-vivo intestinal uptake shows higher uptake of LBL coated MPs than non coated MPs by both the murine macrophage cells (J774) and intestinal epithelial cells (in-vivo).
